Pork Meat Test
TEST PRINCIPLE
Consumption and handling of pork meat is strictly prohibited by some religious confessions. Pork meat may also cause allergy and food intolerance.
Rapid Test (Pork Meat Test) is based on principle of immunochromatographic rapid test (lateral flow) and may be used for qualitative or semi-quantitative determination of pork meat antigen in foods, kitchen and production facilities. In immunochromatographic (lateral flow) method, the target antigens are bound by highly specific antibodies attached to the test line and colored microparticles. This two-site binding results in formation of immune complex visible as the colored line.
TEST SENSIVITY AND SPECIFICITY
In the mixture of meat of different species, Rapid Test (Pork Meat Test) determine at least 1/1000 (ie 0,1%) part of the pork meat antigen. This corresponds approximately to determination of 5-10 milligrams of pork specific antigen per kilogram of the solid material (5-10 ppm). Rapid Test (Pork Meat Test) shows negative result in homogenated meat of following species: cow (beef), sheep, antilopes, horse, moose, reindeer and other deers, chicken, turkey, goose, duck, rabbit, kangaroo. The test is also negative in human blood and muscle tissue.
NOTE: The test gives FALSE NEGATIVE result in meat products processed by long-duration (>3 months) seasoning – e.g. jamon, prosciutto. If the visual test gives unclear results, we recommend to re-check the pork antigen by the quantitative laboratory methods.

Liquid specimens (meat juice, cooked meat brew, washout from kitchen dishes or technological surfaces (eg cutting machines) may be tested directly. The limitation for liquid specimens is their viscosity and the turbidity (presence of particulate material); if the specimen cannot reach the test zone of the strip, it should be diluted by tap water. In this case, the sensitivity level of the test should be adjusted by dilution factor. Turbid specimens should be filtered through textile or paper material, or allowed to set down.
For testing of solid material (fresh or cooked meat, sausages etc.) we recommend the following procedure: using clean (preferably disposable) sharp tool cut a small piece (0.1-0.5 grams) of the material and place it into the Specimen collection tube. Pour 2-2.5 ml of tap water or any other clear water-based liquid (mineral water, soft drink) into the tube; thoroughly close the tube cap and shake the tube vigorously for 15-30 seconds.
- The strips (dipsticks) should be stored at +18oC to +25oC.
- Use the strips within 10 minutes after opening the pouch because the test strips is very sensitive to moisture.
- DO NOT TOUCH the reaction membrane.
- Do not use the strips when its pouch is torn, or test strips is broken or damaged.
- All the components of the test strips are disposable; do not use them repeatedly.
- Do not use the test strips beyond the expiration date
- If the test strips were stored below +20oC, bring the required quantity of the strips to the room temperature before opening the pouches.
- Carefully open the pouch with sharp instrument. TAKE CARE NOT TO DAMAGE THE TEST STRIP.
- Dip the test strip into the liquid part of the specimen observing the right direction and depth (not deeper than the colored line marked by arrow signs) and keep for 10 seconds to ensure soaking.
- Put the test strip onto the CLEAN horizontal surface; do not touch or move the test strip for 10 minutes.
- Read the test result.
Test is considered POSITIVE if TWO colored lines appear in the test zone.
Test is considered NEGATIVE if only ONE coloured line is clearly visible.
If NO colored line is formed, the test is INVALID.
Try to repeat it with another test strip, check the correct specimen handling and test procedure, expiry date and storage conditions.

Pork Meat Test is based on innovative biotechnological product – a pair of monoclonal antibodies against extremely heat resistant component (antigen) of porcine (swine muscle tissue). A target antigen, monoclonal antibodies and immunoassay pair are under international patenting procedure.
The principle of immunochromatography (lateral flow) used in our rapid tests is completely identical to those used for very commonly used pregnancy tests. The only difference is the target substance and specific antibodies.
No, the present version only detects MEAT (muscle tissue). Fat (lard) may give positive result if occasionally contains the rests of muscle tissue.
No, blood is NOT detected. Intestine contains its own muscular tissue and therefore may be positive in present test. We plan to develop separate tests for pork blood, and this test will hopefully detect all internal organs of this animals due to penetration of blood antigens.
No, the test contains only the substances that specifically TARGET and BIND pork, and not pork itself.

As a simple example: the sources of contamination are sometimes a knife previously used to cut the chunks of pork, or the dish washing machine previously used to wash dishes heavily contaminated by pork.
There is NO difference between pork strains and/or origins, a wild pork (boar) is detected equally to domestic pork. A target component (antigen) is EXTREMELY resistant to heat treatment (boiling, roasting, canning) – even heating to 250oC decreases its activity only partly.
NO. This test is not for Halal confirmation or certification, but only to detect the existence of Pork’s protein (not Pork’s DNA) . This test only used for screening and for further verification & detail analysis kindly refer the samples to accredited laboratories.

Use a cotton swab and wipe on the kitchenware. Put the cotton in the specimen tube and filled with water. Kindly follow the instructions on the box.
Pork Meat Test only detect meat antigen (muscle) but can’t be used to detect antigen of the internal organs, blood and pork fat/lard. The level of resistance in meat antigen is very high, more than 200oC higher than the level at which most of foods being processed.
For antigen effect on internal organs, blood & fat/lard need to use other product which is still under development.
